Abstract

Populus deltoides var.'Shalinyang'is an important ecological and economic tree species in the northwest region of China,known for its ecological restoration capabilities and insect resistance.To study the resistance mechanisms the peroxidase(POD)gene PdPOD family which is key secondary metabolite of Populus deltoids on Anoplophora glabripennis(Asian longhorned beetle),the PdPOD gene family was screened and identified based on the transcriptome data of Populus deltoides var.Shalinyang,and was anslyzed by the bioinformatics analysis.And the expression level of the PdPOD gene in phloem of Populus deltoides was detected by qRT-PCR in healthy individuals and those affected by Asian longhorned beetle for 7,18 and 31 d,respectively.The results showed that 42 PdPOD genes were identified,with open reading frames(ORFs)ranging from 327 to 1 134 bp.Phylogenetic analysis classified 42 PdPOD genes into 6 subfamilies,with significant differences in structural features,promoter elements and expression patterns among 6 subfamily members.Additionally,the conserved motifs and protein domains indicated that all 42 members contained the typical domain of plant_peroxidase.The expression patterns of the POD gene family members under insect stress were analyzed based on transcriptome data,the results showed that 19 genes were upregulated.The qRT-PCR results indicated that genes such as PdPOD3 and PdPOD31 exhibited high expression levels under insect stress,suggesting their potential important role in the insect defense response of Populus deltoides var.Shalinyang.Above results not only provided theoretical foundation for understanding the insect resistance functions of PdPOD gene family,but also offered potential genetic resources for the breeding of resistant Populus deltoides var.Shalinyang.
