Development and validation of an UPLC-MS/MS method for the quantification of phillygenin in chicken plasma

Abstract
This study aimed to develop an ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)method for the accurate determination of phillygenin in chicken plasma.Chromatographic separation was performed using an Agilent Poroshell 120 EC-C18 column(100.0 mm×4.6 mm,2.7 µm)with a gradient elution program consisting of methanol and 0.1%formic acid solution as the mobile phase.Mass spectrometric detection was carried out in negative ion mode using multiple reaction monitoring(MRM).The method demonstrated good linearity over the concentration range of 10-1 280 ng/mL(R²>0.99).The intra-day and inter-day precision,expressed as relative standard deviation(RSD),were both below 15%,with accuracy ranging from 92.25%to 106.93%.The mean extraction recovery was between 86.90%and 99.28%,and matrix effects ranged from 89.24%to 104.72%.The samples also showed good stability under all tested storage conditions.The developed method exhibited high specificity,accuracy,and reliable performance,making it suitable for the quantitative analysis of phillygenin in chicken plasma.关键词
