我国首次发现由Alternaria arborescens引起的甘薯叶柄和茎秆枯萎病
First report of leaf petiole and stem blight of sweetpotato caused by Alternaria arborescens in China

Abstract
From 2021 to 2023,sweetpotato samples exhibiting elliptical lesions on leaves,petioles,and stems were collected from Shandong and Yunnan provinces,China.To identify causal agent,pathogens were isolated from diseased tissues and designated as strains Z4hei and YN2,which were identified using morphological and molecular methods.The colonies of both strains were initially white on potato dextrose agar(PDA)and gradually turned dark green to black.Conidia were elliptical or bean-shaped,with 1-6 transverse septa and 0-2 longitudinal septa.The rDNA-ITS sequences of both strains were 569 bp in length and showed 100%identity with each other,exhibiting the highest similarity(100%)with Alternaria alternata(MK370642)and A.arborescens(PP097783)in the NCBI database.To further confirm species identification,the RPB2 and TEF1-α genes were amplified and sequenced.The RPB2 sequences were 985 bp in length and showed 100%identity between the two strains,with 99.59%similarity to A.arborescens(XM_028644303).The TEF1-α sequences were 280 bp in length,sharing 99.60%identity between the strains and showing the highest similarity to A.arborescens(OK081882),with values of 99.29%and 98.93%,respectively.Based on morphological and molecular analyses of rDNA-ITS,RPB2,and TEF1-α sequences,the pathogen causing leaf petiole and stem blight of sweetpotato in China was identified as A.arborescens.This study represents the first report of A.arborescens causing this disease in China.This study provides a theoretical basis for prevention and control of leaf petiole and stem blight of sweetpotato.关键词
