外源褪黑素对干旱胁迫下大豆幼苗生长性能的缓解效应研究
Mitigating Influence of Exogenous Melatonin on Growth Performance of Soybean Seedlings under Drought Stress

Abstract
To clarify the regulatory effects of foliar application of melatonin on the growth of soybean seedlings under drought stress,this study used Suinong 26 as the test variety and simulated drought stress using pot cultivation combined with weight-controlled watering.Water control and melatonin spraying began at the two-leaf stage(designated as Day 0).Two treatments were set up:a drought stress treatment(D,with soil moisture content at 45%-50%of field capacity)and a melatonin treatment under drought stress(D+M,with the same soil moisture content and melatonin application concentration of 100 μmol•L-1 on the same day).A control group(CK)with normal watering and equivalent amounts of distilled water was maintained(soil moisture content at 75%-80%of field capacity).The effects of the melatonin treatment on morphological and physiological traits related to root and above-ground growth in soybean seedlings were analyzed.The results showed that compared to CK,the D treatment significantly inhibited the growth of soybean seedlings.Compared to the D treatment,on Day 20,the D+M treatment increased root length,root volume,and root dry weight by 18.79%,38.15%,and 39.45%,respectively.The increases in root vitality,superoxide dismutase(SOD),peroxidase(POD),catalase(CAT),and ascorbate peroxidase(APX)activities,as well as soluble sugar,soluble protein contents,fructose,and sucrose contents on Days 3,6,and 9 were 33.33%-133.33%,9.74%-8.44%,2.66%-5.73%,27.90%-31.55%,11.81%-16.29%,10.01%-14.07%,11.41%-13.03%,7.71%-11.99%,and 12.66%-21.24%,respectively.Key physiological indicators of photosynthesis,including chlorophyll a,chlorophyll b,total chlorophyll content,carotenoid content,as well as net photosynthetic rate(Pn),transpiration rate(Tr),stomatal conductance(Gs),and intercellular CO2 concentration(Ci)increased by 16.1%-24.4%,4.2%-18.3%,14.9%-23.8%,5.1%-30.5%,22.1%-53.6%,38.7%-99.8%,9.9%-10.4%,and 5.5%-8.1%,respectively,on Days 3,6,and 9.Furthermore,on Day 20,above-ground morphological indicators,including plant height,stem diameter,leaf area,and dry matter accumulation in leaves and stems,as well as growth rate,increased by 22.8%,32.3%,4.3%,25.6%,52.3%,and 18.6%,respectively.Mantel correlation analysis showed that the above-mentioned root morphological and physiological indicators(except for sucrose content)had significant positive correlations with above-ground photosynthetic attributes,dry matter accumulation,and morphological characteristics.Random forest analysis further identified root vitality,root length,and root volume as the three most critical indicators affecting the growth rate of soybean seedlings under drought stress.Thus,it can be concluded that foliar application of melatonin improved key morphological and physiological indicators of both root systems and above-ground parts in soybean seedlings,and there were significant positive correlations between root morphology and key physiological indicators with above-ground growth metrics.关键词
