摘要

Abstract

To address the lack of standard samples for sugarcane yellow leaf virus(SCYLV)nucleic acid detection,this study constructed a recombinant plasmid(pMD19-SCYLV-CP)with the SCYLV coat protein gene,prepared a plasmid DNA standard sample,and verified its characteristics.The results showed that the certified reference material exhibited good homogeneity(F=1.20<F0.05=2.22).Stability assessments revealed that the material remained stable under simulated transport conditions(4-37℃for 14 days),long-term storage(-20℃for 24 months),and repeated freeze-thaw cycles(25 cycles),with consistent collaborative characterization from 12 labs(all SCYLV-positive).This sample provides the first standardized reference for SCYLV detection in China,supporting scientific testing and quality supervision of virus-free seedlings for SCYLV.
