基于流式细胞术和基因组Survey分析的地稍瓜基因组研究
Estimation of Cynanchum thesioides Genome Size Based on Flow Cytometry and Genome Survey Analysis

Abstract
To reveal the chromosome karyotype and genome size of Cynanchum thesioides and provide a reference for whole-genome sequencing,fluorescence in situ hybridization(FISH)was used for karyotype analysis of Cynanchum thesi-oides chromosomes.Using the genome of tomato(Solanum lycopersicum L.)as an internal reference,flow cytometry was employed to estimate the genome size of Cynanchum thesioides.Combined with high-throughput sequencing technology,a genome survey analysis was conducted to evaluate key genome characteristics,including genome size,repetitive se-quence ratio,heterozygosity rate,and GC content.The results showed that the chromosome number of Cynanchum thesioi-des is 2n=2x=24,with chromosome lengths ranging from 0.5 to 1.5 μm,indicating it is a diploid species.The estimated genome size is approximately 163.60~215.04 Mb,with a repetitive sequence ratio of 44.05%,a heterozygosity rate of ap-proximately 1.68%,and a GC content of 34.16%.These findings indicate that the genome of Cynanchum thesioides is a complex genome with high heterozygosity.关键词
