白花泡桐光信号调控基因PfHY5的克隆与功能分析
Cloning and functional analysis of PfHY5 gene in Paulownia fortunei

Abstract
To preliminarily explore the function and transcriptional regulation mechanism of a transcription factor HY5 in Paulownia fortunei,we cloned the coding and promotor sequence of PfHY5 referring to the P.fortunei genome database,followed by bioinformatics and expression analysis.The results showed that the PfHY5 gene was 474 bp in length and encoded 157 amino acids.PfHY5 is a structurally conserved acidic protein located in the nucleus,with its secondary structure mainly composed of α-helices.PfHY5 gene was highly expressed in the stem and root of P.fortunei,and responded to light and Paulownia witches' broom disease.Further,we verified the transcriptional activation activity of its 1993-bp promoter sequence.It was predicted that this region contained cis-elements responsive to light,stress and hormones,indicating that the expression of PfHY5 gene was regulated by multiple factors.These results lay the foundation for clarifying the function of the PfHY5 gene and the light signal transduction pathway in P.fortunei,and also provide a reference for the study of light response mechanism in other woody plants.关键词
