Electroacupuncture Alleviates Neurological Damage in Rats with Acute Cerebral Infarction by Regulating the NLRP3/Caspase-1 Signaling Pathway and Maintaining Blood-Brain Barrier Integrity

Abstract
Objective:To investigate the effects of electroacupuncture(EA)intervention on the NL-RP3/caspase-1 signaling pathway and the expression of blood-brain barrier(BBB)-related factors in rats with acute cerebral infarction,so as to elucidate its potential neuroprotective mechanisms.Methods:A rat model of middle cerebral artery occlusion(MCAO)was established and the rats were randomly divided into a blank control group,model group(MCAO),sham electroacupuncture intervention group(MCAO+sham EA),and electroacupuncture intervention group(MCAO+EA).Cerebral infarction volume was assessed by TTC staining;histopathological changes and neuronal apoptosis were observed by H&E and Nissl staining;neuronal apoptosis,microglial activation,and astrocyte activation were detected by immunofluorescence stai-ning;the expression of neurorepair-related factors was analyzed by immunohistochemistry;and the expression of NLRP3/caspase-1 signaling pathway-related factors as well as BBB tight junction proteins ZO-1 and Clau-din-5 in brain tissue were measured by qRT-PCR and Western blot(WB).Results:EA intervention signifi-cantly reduced the cerebral infarction volume in MCAO rats(P<0.01),alleviated neuroinflammation,and in-hibited neuronal apoptosis.Meanwhile,EA significantly downregulated the expression levels of inflammatory pathway factors such as NLRP3 and caspase-1(P<0.05),and inhibited the overactivation of microglia and astrocytes.Furthermore,EA upregulated the expression levels of tight junction proteins ZO-1 and Claudin-5(P<0.05),mitigated BBB damage,and promoted the expression of neurorepair-related factors such as brain-derived neurotrophic factor(BDNF)and basic fibroblast growth factor(bFGF).Conclusion:Electroacu-puncture may exert anti-inflammatory and neuroprotective effects,thereby alleviating MCAO-induced brain tissue damage,potentially by inhibiting neuroglial cell activation,regulating the NLRP3/caspase-1 signaling pathway,and enhancing blood-brain barrier integrity.关键词
