芥菜型油菜BjB01.MTP5启动子的表达载体构建及表达特性分析

Abstract
In this study,we successfully constructed the promoter expression vector ProBjB01.MTP5-GUS for BjB01.MTP5 in Brassica juncea.Phylogenetic and cis-acting element analyses were performed based on the promoter sequences of the MTP5 gene from ten Brassicaceae species.The results revealed that the MTP5 promoters of the B subgenome in Brassica formed distinct evolutionary branches compared with those of the A/C subgenomes.Analysis of cis-acting elements indicated that elements responsive to phytohormones(e.g.,abscisic acid,methyl jasmonate,and auxin)and abiotic stresses including anaerobic induction,salicylic acid,drought,and low temperatures were highly conserved in the MTP5 promoters across different species.Tissue-specific expression profiling demonstrated that BjB01.MTP5 was predominantly expressed in roots,flower buds,siliques,and pericarps.Furthermore,qRT-PCR analysis under heavy metal stress showed that the expression of BjB01.MTP5 was significantly induced by various heavy metals,such as Mn2+,Fe2+,Zn2+,and Sb3+.Its expression pattern correlated strongly with the distribution of heavy metal-responsive elements in the promoter region.These findings provided important theoretical insights into the biological functions of MTP genes in response to heavy metal stress.关键词
