阿魏酸对H2O2诱导的猪肠道上皮细胞氧化损伤的保护作用

Abstract
To elucidate the underlying mechanisms of ferulic acid(FA)protects against hydrogen peroxide(H2O2)-induced oxidative damage,this study compared the differences in the types and content of phenolic acids such as ferulic acid(FA)in the whole and peeled fruit juice samples of Gannan navel oranges.Using the H2O2 treatment group as the control,the effect of FA treatment on porcine intestinal epithelial cells(IPEC-J2)were analyzed,including:superoxide dismutase(SOD)activity,catalase(CAT)activity,malondialdehyde(MDA)content,reactive oxygen species(ROS)levels,cell morphology,cell viability,lactate dehydrogenase(LDH)leakage rate,and mitochondrial membrane potential.The results revealed that the content of FA in the whole fruit juice was 5.8 times of that in the peeled fruit juice.The results showed that the content of FA in the whole fruit juice was 5.8 times that in the peeled fruit juice.FA markedly ameliorated H2O2-induced cell vitality loss and apoptosis in IPEC-J2.The protective efficacy of FA might be ascribed to the increases in superoxide dismutase(SOD)and catalase(CAT)activities and the decreases in lactate dehydrogenase(LDH)release,reactive oxygen species(ROS),and malondialdehyde(MDA)in IPEC-J2 cells.Meanwhile,our study revealed correlations between mitochondrial Cyt-c-mediated caspase-3 activation pathway and the antioxidant effect of FA on IPEC-J2.Furthermore,FA reduced the activation of p53 and caspase-8 mediated by H2O2.Taken together,our results indicated that FA protected IPEC-J2 cells against H2O2-mediated oxidative stress and cell injury by resistance to apoptosis via the inhibition of p53-and caspase-3-dependent signaling pathways.The current study indicated that FA could serve as a potentially effective candidate medicine for the treatment of intestinal oxidative damage.关键词
