Separation of antioxidants from Nigella glandulifera seeds using pH-responsive deep eutectic solvents

Abstract
To develop a green and efficient extraction technology for natural products,a pH-responsive DES(deep eutectic solvent)system was constructed for extracting antioxidant components from Nigella glandulifera seeds.The optimal DES system was selected based on antioxidant activity assessment,followed by process optimization through Box-Behnken design.FT-IR(fourier transform infrared spectroscopy)and 1H NMR(1H nuclear magnetic resonance)analysis were employed to characterize intermolecular hydrogen bonding in DES systems,while SEM(scanning electron microscopy)revealed microstructural changes on the sample surface before and after extraction.The results demonstrate that using a ternary DES system composed of ethanolamine,octanoic acid and water(molar ratio 1∶1∶4)as the extraction solvent,the antioxidant activity is the highest,and the value of ferric reducing antioxidant power is(35.76±0.98)μmol/g,under optimized conditions of extraction temperature 51 ℃,liquid-solid ratio 29∶1 mL/g,extraction time 26 min and phase transition initiator dosage(volume ratio)5∶1.FT-IR and 1H NMR analysis confirm the formation of cyclic structures through hydrogen bonding between DES components.SEM observations reveal that the optimized DES system significantly enhances mass transfer efficiency by disrupting cellular walls.This pH-responsive DES system shows superior extraction performance for Nigella glandulifera antioxidants compared with conventional solvents,which provides technical reference for developing sustainable separation technologies in natural product extraction.关键词
