摘要

Abstract

The correlation between the active components(alkaloids)of Fritillaria ussuriensis plant and their key enzyme genes in the alkaloid synthesis was studied for providing a basis for revealing the synthesis mechanism.The alkaloid content of samples from 17 origins was detected by HPLC-MS,and the expression levels of HMGR,FPS and DXR genes regulating alkaloid synthesis in each sample were determined by qPCR using 18S rRNA as an internal reference gene.The correlation be-tween alkaloid content and gene expression was studied by bivariate correlation analysis method.The results showed that in leaves,HMGR and FPS genes were negatively correlated with the content of peimine,peiminine and total alkaloids,DXR gene was significantly positively correlated with pei-mine(P<0.05),and positively correlated with peiminine and total alkaloids.In bulbs,HMGR gene was negatively correlated with peimine,and positively correlated with peiminine and total alkaloids,FPS gene was significantly negatively correlated with peimine(P<0.05),and negatively correlated with peiminine and total alkaloids,DXR gene was significantly negatively correlated with peimine,peiminine and total alkaloids(P<0.05).Therefore,it is inferred that HMGR,FPS and DXR genes have regulatory effects on alkaloid synthesis,and alkaloid accumulation is more obviously regulated by DXR in the MEP pathway,but not obviously regulated by HMGR and FPS in the MVA pathway.
