小青龙汤联合虫类药及地塞米松对哮喘豚鼠的治疗作用及其机制研究
Therapeutic effect and mechanism of Xiaoqinglong Decoction combined with insect drugs and dexamethasonein guinea pigs with asthma

Abstract
Objective To investigate the therapeutic effect and mechanism of Xiaoqinglong De-coction combined with insect drugs(Pheretima,Bombyx Batryticatus,Scorpio)and western medicine(dexamethasone)on guinea pigs with asthma based on the signal transducer and activator of tran-scription 3(STAT3)/transforming growth factor-β1(TGF-β1)signaling pathway.Methods Fifty male guinea pigs were randomly divided into normal control group,model group,western medicine group(treated with dexamethasone),traditional Chinese medicine group(treated with Xiaoqinglong Decoction combined with insect drugs),and combined treatment group(treated with dexamethasone combined with Xiaoqinglong Decoction and insect drugs),with 10 guinea pigs in each group.Except for the normal control group,chronic asthma models were established in the other groups,and the respective treatment groups were administered the corresponding drugs by gavage.Airway resistance and pulmonary dynamic compliance were measured in each group.Pulmonary tissue pathological morphology was observed,and semi-quantitative analysis was conducted in indicators related to airway inflammatory infiltration,airway wall thickness,and collagen deposition.Serum interleukin(IL)-6,IL-13,and TGF-β1 concentrations were determined,and the expressions of phosphorylated STAT3(p-STAT3),STAT3,TGF-β1,and α-smooth muscle actin(α-SMA)in pulmonary tissues were detected.Results Compared with the model group,the airway resistance decreased and the pulmo-nary dynamic compliance increased in the western medicine group,the traditional Chinese medicine group,and the combined treatment group,with statistically significant differences(P<0.05).The airway resistance in the combined treatment group was lower than that in the western medicine group and the traditional Chinese medicine group,and the pulmonary dynamic compliance was higher than that in the western medicine group and the traditional Chinese medicine group,with statistically sig-nificant differences(P<0.05).The airway wall thickness and the proportion of collagen area in the western medicine group,the traditional Chinese medicine group,and the combined treatment group were smaller than those in the model group,and those in the combined treatment group were smaller than those in the western medicine group and the traditional Chinese medicine group,with statisti-cally significant differences(P<0.05).The serum levels of IL-6,IL-13,and TGF-β1 in the west-ern medicine group,the traditional Chinese medicine group,and the combined treatment group were lower than those in the model group,and those in the combined treatment group were lower than those in the western medicine group and the traditional Chinese medicine group,with statistically significant differences(P<0.05).The relative protein expressions of p-STAT3/STAT3,TGF-β1,and α-SMA in the western medicine group,the traditional Chinese medicine group,and the com-bined treatment group were lower than those in the model group,and those in the combined treat-ment group were lower than those in the western medicine group and the traditional Chinese medicine group,with statistically significant differences(P<0.05).Conclusion Xiaoqinglong Decoction combined with insect drugs and dexamethasone has a synergistic effect in treating guinea pigs with asthma.The mechanism may be related to the synergistic regulation of the STAT3/TGF-β1 signaling pathway,which can inhibit airway inflammation through multiple links and effectively delay the process of airway remodeling.关键词
