Prognostic value of NR3C2 and immune infiltration analysis in glioma

Abstract
Objective To explore the prognostic value of NR3C2 in glioma and its relationship with the immune infiltration landscape.Methods Based on RNA-seq expression data from glioma patients in The Cancer Genome Atlas(TCGA)database,the expression characteristics of NR3C2 were systematically analyzed.The Kruskal-Wallis rank-sum test was used to assess the associations between NR3C2 expression and clinicopathological features of glioma,including age,isocitrate dehydrogenase(IDH)status,1p/19q codeletion,and World Health Organization(WHO)grade.The expression levels of NR3C2 in glioma cells were validated by reverse transcription quantitative PCR(RT-qPCR),Western blotting,and immunofluorescence assays.Kaplan-Meier survival analysis was performed to evaluate the association between NR3C2 expression and overall survival(OS),while receiver operating characteristic(ROC)curve analysis was used to assess its predictive performance.Immune cell infiltration levels were estimated using the single-sample gene set enrichment analysis(ssGSEA)algorithm,and Spearman's rank correlation analysis was performed to examine the relationship between NR3C2 expression and immune cell infiltration.Results NR3C2 expression was significantly down-regulated in glioma tissues and showed significant associations with WHO grade,IDH status,age,and 1p/19q codeletion.Lower NR3C2 expression was associated with poor prognosis and reduced survival duration.Experimental assays further demonstrated that NR3C2 expression in glioma cells was significantly decreased compared with that in normal astrocytes.Immune infiltration analysis indicated significant correlations between low NR3C2 expression and the infiltration of neutrophils,T cells,and macrophages.Conclusions NR3C2 is expressed at low levels in glioma,and its expression is closely associated with patient prognosis and immune cell infiltration,suggesting that NR3C2 may be involved in the regulation of the glioma immune microenvironment and play a potential role in tumor immune modulation.关键词
