葛根素/液态金属水凝胶的制备及其介导三阴性乳腺癌光热治疗研究
Preparation of Puerarin/Liquid Metal Hydrogel and Its Application in Photothermal Therapy of Triple-Negative Breast Cancer

Abstract
Objective To construct a novel puerarin/liquid metal(Pue/LM)hydrogel for the photothermal therapy of triple-negative breast cancer(TNBC).Methods A stable Pue/LM hydrogel was fabricated by optimizing the ratio of Pue to LM.Its structural characteristics and properties were systematically characterized using scanning electron microscopy(SEM),energy-dispersive spectroscopy(EDS),Fourier transform infrared spectroscopy(FTIR),X-ray diffraction(XRD),and ultraviolet-visible spectroscopy(UV-vis).The photothermal conversion efficiency was calculated based on temperature changes under 1064 nm near-infrared(NIR)light irradiation,and photothermal response stability was verified via cyclic heating-cooling experiments.In vivo photothermal efficacy was evaluated using animal models.Additionally,live/dead cell staining,flow cytometry,and mitochondrial membrane potential detection were combined to investigate the photothermal killing effect and underlying mechanism of Pue/LM on 4T1 cells under NIR light irradiation.Results The Pue/LM hydrogel was successfully constructed with optimized proportions,achieving efficient loading of LM.Under 1064 nm NIR light irradiation,the hydrogel exhibited significant photothermal conversion capacity and excellent cyclic stability.In vivo experiments confirmed its photothermal response at the tumor site.Cellular studies demonstrated that,under NIR light irradiation,the Pue/LM hydrogel remarkably increased the late apoptosis and necrosis rates of 4T1 tumor cells by reducing mitochondrial membrane potential and disrupting mitochondrial function.Conclusions This study successfully developed a novel Pue/LM hydrogel,which shows promising therapeutic potential for TNBC under photothermal conditions,providing a new strategy for the local treatment of this disease.关键词
