摘要

Abstract

[Objective]This study aimed to investigate the adhesion target of Paranosema locustae and its mediated toxicity mech-anisms.[Methods]Bioinformatic analysis and quantitative real-time PCR(qPCR)were employed to examine the molecular characteristics and expression patterns of a hypothesized adhesion gene(ADH1)in P.locustae.[Results]The cloned open reading frame of the putative adhesion gene was 885 bp,encoding 294 amino acids with a molecular weight of approximately 32.73 kDa and an isoelectric point of 8.69.Spatiotemporal expression analysis revealed significant temporal and spatial specific-ity:temporally,the ADH1 gene was highly expressed across tissues during both early(3 days post-infection,dpi)and late(12~18 dpi)infection stages,while during the mid-stage(6~9 dpi),expression focused on tissues associated with pathogen colonization and immunity,such as the gonads and Malpighian tubules;spatially,the tissues with high expression dynamically shifted over the course of infection:early expression concentrated in the hindgut,mid-stage expression focused on the gonads,and late-stage expression was also prominent in immune-related tissues such as the fat body and Malpighian tubules.Notably,in all highly expressed tissues,the transcriptional peak of ADH1 preceded the increase in P.locustae pathogen load.[Conclu-sion]It was confirmed that ADH1 played a critical role in the infection of host cells by P.locustae,and that adhesion may occur prior to spore activation and host cell infection.This study provided a new perspective into the infection mechanism of P.locus-tae and laid a foundation for further research into the pathogenic process.
