Abstract

In this study,traditional isolation and culture-based methods were used to characterize the diversity and community structure of endophytic microorganisms(bacteria and fungi)in the stems and leaves of Angelica sinensis from three major production regions in Gansu province,China:Zhangxian(ZX),Minxian(MX),and Tanchang(TC)counties.The results showed that a total of 435 endophytic bacterial strains,belonging to 35 genera and 108 species,and 297 endophytic fungal strains,belonging to 15 genera and 89 species,were isolated from the stems and leaves of A.sinensis.The richness and diversity of both endophytic bacteria and fungi were the highest in the leaves from the TC region,whereas the diversity of the bacterial community in the stems and the fungal community in the leaves from the ZX region was the lowest.The dominant endophytic bacterial genera in A.sinensis from vari-ous habitats were Bacillus,Pseudomonas and Lysinibacillus.Bacillus was enriched in the leaves of the ZX region,while Pseudomonas was dominant in the TC leaves.The dominant fungal genera were Fusarium,Alternaria,and Calophoma.Fusarium was consistently enriched in the stems across all three habitats,while Alternaria was predominated in the leaves of the ZX and MX regions.Pearson correlation analysis indicated that the bacterial community structures in the stems exhibited relatively weak correlations at the genus level among different habitats,whereas the fungal communities showed a highly significant correlation(P<0.001).In addition,a significant positive correlation(P<0.05)was observed in the endophytic bacterial communities of leaves from the MX and TC plots,while an extremely significant positive correlation(P<0.001)was found in the endophytic fungal communi-ties of the leaves from the ZX and MX plots.Principal component analysis(PCA)and correspondence analysis(CA)indicated that both habitat differences and tissue-specificity jointly influenced the struc-ture of endophytic microbial communities in the stems and leaves of A.sinensis.
