Abstract

Objective To explore the pharmacological basis and mechanism of action of Buyang Huanwu Decoction in improving the pain threshold of diabetes mellitus(DM).Methods Firstly,the effect of Buyang Huanwu Decoction on the pain threshold of DM model rats was observed.Then,network pharmacology and molecular docking techniques were used to screen the potential active components and key targets of the decoction in improving the pain threshold of DM.Results Buyang Huanwu Decoction could increase the body weight of DM rats,lower their blood glucose levels,and significantly enhance the pain threshold.A total of 104 active components of the decoction for pre-venting diabetic foot(DF)were screened,among which quercetin,kaempferol,and stigmasterol were the core active components.A total of 18 important targets were screened,and the first five key targets were IL6,IL1B,TNF,STAT3 and AKT1.Molecular docking indicated that the binding energies of quercetin,kaempferol and TNF,stigmasterol and IL6,and quercetin and IL1B were all lower than-5 kcal/mol,sug-gesting good binding activity.Conclusion Buyang Huanwu Decoction can effectively increase the mechanical and thermal pain thresholds of DM rats,providing new experimental evidence for explaining the comprehensive pharmacological mechanism of the formula in preventing DF pain thresholds through a"multi-component,multi-target"approach.
