Comparison of diethylnitrosamine-induced primary liver cancer models in mice at different developmental stages

Abstract
Objective To systematically compare the differences in tumorigenic efficiency,pathological characteristics,and molecular phenotypes between neonatal(14 d post-birth)and adult(7 weeks old)C57BL/6 mouse models of diethylnitrosamine-induced hepatocellular carcinoma(HCC).Methods Thirty wild-type C57BL/6 mice were selected and divided into a normal group,a neonatal group,and an adult group,with 10 mice in each group.The induced HCC mouse model was established by intraperitoneal injection of carbon tetrachloride combined with a high-fat diet.The survival rates,liver histomorphology,serum liver function indicators[alpha-fetoprotein(AFP),alanine aminotransferase,aspartate aminotransferase,total bilirubin,direct bilirubin,albumin,and triglyceride],pathological staining(HE,PAS,and ORO),and immunohistochemical staining were dynamically monitored to observe the expression and distribution of Ki67 and AFP.Results The pathological features of liver tissues and serum AFP levels(P<0.01)in the neonatal group more closely resembled the clinical progression pattern of HCC.In contrast,the adult group exhibited a prolonged latency period and milder metabolic disturbances.Conclusion This study provides critical experimental evidence for the selection of induced HCC animal models and advances research on developmental stage-dependent carcinogenic mechanisms.关键词
