Abstract

Objective To verify the therapeutic effect of growth differentiation factor 15(GDF15)neutralizing monoclonal antibody on glioblastoma(GBM)and to confirm the feasibility of GDF15 as an immunotherapy target for GBM.Methods The therapeutic efficacy of GDF15 neutralizing antibody was evaluated by using orthotopic GBM model mice and spontaneous GBM model mice.Additionally,the combined therapeutic effects of GDF15 neutralizing antibody and PD-1 monoclonal antibody were investigated,and their clinical application prospect in the treatment of GBM was assessed.Results GDF15 neutralizing antibody demonstrated significant efficacy in inhibiting glioma progression and effectively prolonged survival in both orthotopic GBM and spontaneous GBM mouse models.Moreover,the combined therapy of GDF15 neutralizing antibody and PD-1 monoclonal antibody exhibited superior anti-tumor effects compared to monotherapy with either antibody alone.Conclusion GDF15 neutralizing monoclonal antibody effectively inhibits glioma growth,likely by reversing tumor-induced immunosuppression and enhancing anti-tumor immune responses.This study confirms the potential application value of GDF15 as a target for GBM immunotherapy to a certain extent and provides a direction for the selection of new targets and intervention strategies for GBM immunotherapy.
