PP2对新城疫病毒体外抑制作用的研究
Inhibitory effect of PP2 on Newcastle disease virusin vitro

Abstract
To evaluate the impact of the Src family tyrosine kinase(SFKs)inhibitor PP2 on Newcastle disease virus(NDV)infection,this study used chicken macrophage HD11 cells as a model and determined the working concentration of PP2 in HD11 cells by assessing cell viability.Subsequently,indirect immunofluorescence,Western-blot,reverse-transcription fluorescent quantitative PCR(RT-qPCR)and hemagglutination assays were employed to assess the effects of PP2 treatment on different stages of NDV infection.The results showed that PP2 significantly inhibited protein expression and progeny virus release of NDV in HD11 cells.Further investigation revealed that PP2 did not affect the adsorption and entry process of NDV,but suppressed the RNA synthesis step that occurred after viral entry.Moreover,PP2 exhibited anti-NDV activity in multiple cell types.Additionally,the SFKs inhibitors PP1 and RK24466,which share the same target as PP2,exhibit comparable anti-NDV efficacy in vitro.In summary,PP2 could effectively inhibit NDV replication.This study provides insights for host-targeted antiviral strategies against NDV.关键词
