Potential value of guanylate binding protein 1 in prognosis and immune regulation of pan-cancer

Abstract
Objective To explore the expression pattern,prognostic significance and immune regulatory role of guanylate binding protein 1(GBP1)in pan-cancer.Methods The expression of GBP1 gene and protein in pan-cancer was analyzed based on The Cancer Genome Atlas(TCGA),the University of Alabama at Birmingham cancer data analysis portal(UALCAN)and the Human Protein Atlas(HPA)database.The TISIDB database and R software were used to investigate the relationship between GBP1 and molecular subtypes,pathological stages and prognosis of tumor patients.The correlation between GBP1 and tumor functional status could be analyzed by STRING,GSCALite and cancerSEA databases.The relationship between GBP1 expression and tumor mutation burden(TMB),mutant allele tumor heterogeneity(MATH),tumor purity,microsatellite instability(MSI),ploidy,neoantigen(NEO),homologous recombination defect(HRD)and heterozygosity loss(LOH)was detected by Pearson correlation analysis.The correlation between GBP1 and immune regulation was analyzed based on R software,TCGA and SpatialDB databases.The effects of GBP1 knockdown on the proliferation,migration and invasion of pancreatic cancer cells were investigated in vitro.Results GBP1 was abnormal in many cancer types and had high diagnostic accuracy.GBP1 expression was significantly different in different molecular subtypes and pathological stages of various human cancers,and GBP1 expression is correlated with the prognosis of patients with various tumors.GBP1 expression was associated with DNA damage,hormone AR,apoptosis,epithelial-mesenchymal transformation,inflammation,and tumor stemness.GBP1-related genes were enriched in focal adhesion,IL10 synthesis,PD1 signaling pathway,MAPK activation,Th17 cell differentiation and other pathways.GBP1 was correlated with tumor purity,TMB,MSI,ploidy,MATH,NEO,HRD and LOH in pancarcinoma.The expression of GBP1 was related to immune subtype,immune score and immune cell infiltration.GBP1 knockdown significantly inhibited the proliferation,migration and invasion of pancreatic cancer cells.Conclusions GBP1 has the potential to be a biomarker for the diagnosis,prognosis and immunotherapy of tumor patients.关键词
