Abstract

This study employed tandem mass tag(TMT)-labeled proteomics technology to investigate the changes in protein expression in the hepatopancreas of Litopenaeus vannamei under 0.2 mg/L Cd2+stress for 6 d,aiming to elucidate the Cd accumulation and tolerance mechanisms in typical marine crustaceans.The results revealed a total of 369 differential proteins in the stress group compared with the control group,including 214 upregulated and 155 downregulated proteins.The gene ontology(GO)analysis indicated that these differential proteins were primarily involved in biological processes such as cellular process and metabolic process,cellular components including cells and protein-binding complexes,and molecular functions dominated by catalytic activity and binding.The eukaryotic orthologous groups(KOG)annotation showed significant enrichment in functions such as post-translational modification and energy metabolism.Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis demonstrated that upregulated proteins were associated with the protein processing in endoplasmic reticulum pathway,while downregulated proteins were concentrated in ribosome-related pathways,as well as arginine and proline metabolism pathways.Under the Cd stress,key proteins such as zinc finger proteins,metallothionein-like,calnexin,calreticulin,heat shock proteins(HSPs)family(HSP60,HSP70,and HSP90),and antioxidant enzymes like glutathione peroxidase(GPx)were significantly upregulated.These proteins may collectively form a molecular response network in Litopenaeus vannamei to counteract Cd2+stress by regulating Cd2+binding and efflux,repairing misfolded proteins,alleviating endoplasmic reticulum stress,and scavenging reactive oxygen species.
