Development of Markers for Specific Identification of the Heat-tolerant Gene TT2 in Rice

Abstract
This study developed high-efficiency molecular identification markers for the heat-tolerant gene TT2 in rice,so as to accelerate the precise screening of heat-tolerant germplasm resources and heat-tolerant variety breeding of rice.According to the basic principle of amplification of amplification refractory mutation system PCR(ARMS-PCR)and the single nucleotide polymorphism(SNP)differences within the TT2 gene,three sets of specific primers were designed for the agarose gel electrophoresis,capillary electrophoresis,and penta-primer amplification refractory mutation system(PARMS)technology,respectively,enabling accurate discrimination of different TT2 genotypes.The agarose gel electrophoresis marker identified TT2 genotypes by the amplicon size,with high accuracy and simple operation.The capillary electrophoresis marker distinguished TT2 genotypes via the fluorescent fragment size,offering high throughput and speed.The PARMS marker achieved high-throughput and precise TT2 genotyping via a microplate reader,which was faster,simpler and more convenient.These three molecular marker systems with different detection methods can meet the detection requirements of laboratories with varied conditions,allowing the accurate identification of elite TT2 haplotypes.They provide key technical tools for pyramiding heat-tolerant genes and breeding new heat-tolerant varieties of rice,thereby promoting the development of heat-tolerant rice breeding.关键词
