Abstract

The secure preservation of germplasm resources serves as a critical foundation for crop breeding,and developing efficient conservation methods for weak-growth accessions is particularly crucial.This study utilized 135 accessions of weak sweet potato seedlings as experimental materials.These materials were hydroponically cultured for 21 d to induce rooting,subsequently transplanted into potting substrate for continued conservation,with their post-transplant growth recovery systematically monitored.The results demonstrated that hydroponic treatment induced robust rooting in weak seedlings,with 78.5%of accessions(line)producing 4-18 roots per plant and 88.1%exhibiting an average root length of 0.6-3.1 cm.At 20 d post-transplantation,the overall survival rate recorded 82.2%.After five months,96%of the accessions(line)produced vine lengths longer than 30 cm,A significant increase was observed in basal branching number,while 91%attained stem diameters greater than 3.0 mm,confirming that the overall growth met the established criteria for successful rejuvenation.Virus elimination was subsequently applied to the hydroponically-rejuvenated accessions(line),achieving a success rate of 90.1%.Virus elimination significantly restored the tuberization capacity in conventional seedlings of non-tuberizing accessions.The results showed that 82.4%of accessions(line)with virus-free seedlings achieved a yield exceeding 1000 kg per mu,whereas only 27.5%of conventional seedlings from the same accession reached equivalent production levels.The hydroponic rejuvenation technique not only enhanced the survival capacity of weak germplasm seedlings but also secured a critical time window for subsequent virus elimination,thereby providing a reliable and viable approach for the rejuvenation and conservation of weak seedlings within sweet potato germplasm resources.
