Shenyuan Granules Improve Cognitive Function in Chronic Kidney Disease Rats by Regulating Klotho/NF-κB Pathway

Abstract
Objective To investigate the effects of Shenyuan Granules(SYG)on inflammatory response and cognitive function in rats with chronic kidney disease(CKD)based on the Klotho(KI)/nuclear factor-κB(NF-κB)signalling pathway.Methods A CKD model was established in rats via 5/6 nephrectomy.The rats were divided into sham-operated group,model group,SYG group,and positive drug group,with 6 rats in each group.The SYG group was administered SYG(6.0 g/kg)by gavage once daily,while the positive drug group received donepezil hydrochloride(10 mg/kg).The sham-operated and model groups were given an equal volume of saline.Cognitive function was assessed using the Morris water maze test.Serum levels of urea nitrogen(BUN)and creatinine(Cr)were measured biochemically.Serum concentrations of TNF-α and IL-1β were detected by ELISA.Protein expression of KI and NF-κB in the kidney and brain was determined by Western Blot.Pathological changes in the hippocampus and kidney were observed under light microscopy.Results Compared with the sham-operated group,the model group exhibited prolonged escape latency(P<0.01),reduced number of platform crossings and duration in the target quadrant(P<0.0001),elevated serum BUN and Cr levels(P<0.0001),increased TNF-α and IL-1β concentrations(P<0.0001),decreased KI protein expression in kidney and brain tissues(P<0.0001),and increased NF-κB protein expression(P<0.0001).Compared with the model group,both the SYG and positive drug groups showed shortened escape latency(P<0.001),increased platform crossings(P<0.05),prolonged platform stay(P<0.01),decreased BUN and Cr(P<0.05,P<0.01,P<0.0001),reduced TNF-α and IL-1β concentrations(P<0.0001),upregulated KI protein expression(P<0.001,P<0.0001),and downregulated NF-κB protein expression(P<0.001,P<0.0001))in kidney and brain.No significant pathological changes were observed in the sham-operated group.The model group showed marked pathological changes in the hippocampus and kidney,which were ameliorated in the SYG and positive drug groups.Conclusions SYG may alleviate inflammation and improve cognitive function in CKD rats,potentially by regulating the KI/NF-κB pathway.关键词
