温胆汤对脂多糖诱导的RAW264.7巨噬细胞极化及迁移的影响
Regulatory effects of the Wendan decoction on lipopolysaccharide-induced polarization and migration in RAW264.7 macrophages

Abstract
Objective:To investigate the effects of serum containing the Wendan decoction(温胆汤)on polarization and migration of RAW264.7 macrophages induced by lipopolysaccharide(LPS).Methods:M1-type polarization model was established by stimulating RAW264.7 macrophages with LPS.Cell viability was assessed using cell counting kit-8(CCK-8)assay to screen the optimal concentration of serum containing Wendan decoction for intervention in macrophages.Subsequently,macrophages were treated with the serum containing Wendan decoction,and their migration ability was evaluated via transwell migration assays and scratch wound assays.The levels of interleukin-10(IL-10)and interleukin-6(IL-6)were measured by ELISA,while mRNA expression of IL-6,iNOS(M1 markers),IL-10,and Arg-1(M2 markers)was quantified using RT-qPCR.Results:Medicated serum containing Wendan decoction can reduces the migration number of RAW264.7 macrophages induced by LPS and increases the scratch closure distance of macrophages(P<0.01).It can increase the levels of IL-10 in the macrophage supernatant(P<0.05)and decrease the levels of IL-6(P<0.05),downregulate the expression of mRNA of IL-6 and iNOS(M1 polarization markers)(P<0.05),upregulate mRNA expression of IL-10 and Arg-1(M2 polarization markers)(P<0.05).Conclusion:Medicated serum containing Wendan decoction could inhibit macrophage migration and M1 polarization induced by LPS,thereby suppressing inflammatory responses.The study suggests its potential as a therapeutic agent for inflammation-related diseases.关键词
