整合素-α2对人根尖牙乳头干细胞增殖、迁移及成骨分化的体外调控初步研究
Preliminary study on the regulation of integrin-α2 on proliferation,migration and osteogenic differentiation of human stem cells from apical papilla in vitro

Abstract
Objective To initially explore the in vitro regulatory effect of integrin-α2(ITGA2)on the prolifera-tion,migration and osteogenic differentiation of human stem cells from apical papilla(SCAP).Methods SCAPs stably silenced by Lentivirus were constructed.Set the ITGA2 silencing group as ITGA2(-),and the empty vec-tor control group as Control.The effect of ITGA2 on SCAP proliferation ability was evaluated by CFU-F to detect colony formation rate and CCK-8 to detect growth curve,migration ability was evaluated by scratch test and Tran-swell test,apoptosis rate was analyzed by FCM,and changes in apoptotic genes and stem genes were detected by RT-qPCR,After osteogenic induction,ALP staining,ARS staining and quantitative analysis of calcified nodules were used to evaluate their effects on the osteogenic differentiation ability of SCAP.Results After silencing IT-GA2,the proliferation and migration ability of SCAP was significantly reduced(P<0.05).While the flow cy-tometry apoptosis rate increased on average,RT-qPCR results showed that the expression of genes that inhibit ap-optosis(BCL2)and stem genes(OCT4)were reduced(P<0.05).After osteogenic induction,the staining in-tensity of the ITGA2(-)group was lighter than that of the Control group.Semi-quantitative analysis of calcified nodules also showed that the ITGA2(-)group was less mineralized(P<0.05).Conclusion Silencing ITGA2 can inhibit the proliferation,migration and osteogenic differentiation of SC AP in vitro,and also affect the apopto-sis process and stemness gene expression of SCAP.关键词
