球花石斛离体快繁技术研究
In Vitro Rapid Propagation Technology of Dendrobium thyrsiflorum

Abstract
Taking capsules of Dendrobium thyrsiflorum as research materials,this study explored their morphological char-acteristics and seed germination traits,screened the optimal medium for seed germination,and conducted systematic and in-depth research on protocorm proliferation as well as strong seedling cultivation and rooting to establish an in vitro rapid propagation technology system.The system is expected to be applied to germplasm resource conservation and better serve industrialized seedling raising.The results showed that the capsules of Dendrobium thyrsiflorum were long oval,with an av-erage length of 2.14 cm and a central diameter of 1.05 cm;the longitudinal surface of the capsule was roughly triangular,and the pericarp at the ventral suture slightly protruded outward.The seeds were golden and spindle-shaped,with a thin membra-nous seed coat and an approximately oval embryo;the average seed length was 327.46 μm,with a central diameter of ap-proximately 84.73 μm.The highest germination rate of 86.96%was achieved when seeds were inoculated on M8 medium(MS+60 mL·L-1 banana juice).The protocorm proliferation rate reached the highest value of 201.35%on K9 medium(2/3 MS+60 mL·L-1 coconut water+0.4 mg·L-1 6-BA).Based on the comprehensive analysis of three indicators,including root number,root length,and plant height,Dendrobium thyrsiflorum performed the best in rooting on G3 medium(MS+60 mL·L-1 banana juice+0.6 mg·L-1 NAA+0.5 g·L-1 AC),with an average root number of 15.2,an average root length of 3.3 cm and an average plant height of 4.58 cm.关键词
