The Effect of Cinnamaldehyde on Estrogen Secretion and Apoptosis of Mouse Ovarian Granulosa Cells

Abstract
Ovarian granulosa cells(GCs)of mice are the core cells responsible for estrogen secretion.This study aimed to investigate the effect of cinnamaldehyde treatment on estrogen secretion and apoptosis of mouse ovarian GCs.In this study,3-week-old SPF-grade female KM mice were selected.The mouse ovarian GCs were isolated and divided into a blank control group,a 5× 10-5 mol·L-1 cinnamaldehyde treatment group,and a 10×10-5 mol·L-1 cinnamaldehyde treatment group.Each group had 3 replicates.Firstly,this study analyzed the gene expression profiles of cinnamaldehyde-treated mouse follicular GCs and the enrichment of differentially expressed genes(DEGs)using transcriptome sequencing analyses.Then,ELISA,cell counting Kit-8(CCK-8),flow cytometry,and RT-qPCR were employed to clarify the effect of cinnamaldehyde on estrogen secretion and apoptosis of mouse GCs.The sequencing results showed that the DEGs in mouse ovarian GCs treated with 5×10-5 mol·L-1 cinnamaldehyde were enriched in the PI3K-Akt signaling pathway,tumor-related pathways,AGE-RAGE signaling pathway in diabetic complications,and progesterone-mediated oocyte maturation,among others.The experimental results showed that treatment of mouse ovarian granulosa cells GCs with 5×10-5 mol·L-1 cinnamaldehyde could promote estrogen secretion by significantly upregulating the relative mRNA expression levels of StAR,CYP19A1,and CYP11A1.Compared with the control group,treatment with 5×10-5 mol·L-1 and 10×10-5 mol·L-1 cinnamaldehyde both induced apoptosis of mouse ovarian GCs to different degrees.The 5×10-5 mol·L-1 cinnamaldehyde treatment significantly upregulated the relative mRNA expression levels of CHOP,IRE1,ATF6,and PERK,while 10×10-5 mol·L-1 cinnamaldehyde treatment significantly upregulated the relative mRNA expression levels of CHOP and ATF6.In conclusion,5×10-5 mol·L-1 cinnamaldehyde may promote estrogen secretion in mouse ovarian GCs by upregulating the expression of steroid hormone synthases via the PI3K-Akt signaling pathway.Additionally,both 5×10-5 mol·L-1 and 10×10-5 mol·L-1 cinnamaldehyde can induce GCs apoptosis through the endoplasmic reticulum stress(ERS)pathway.关键词
