Abstract

[Objective]Meloidogyne hapla affects the yield and quality of Chinese medicinal materials.Predicting the secreted proteins of M.hapla and designing specific primers lay a foundation for the rapid and accurate identification of M.hapla.[Method]SignalP 6.0,TargetP-2.0,WoLF PSORT,ProtComp 9.0,DeepTMHMM1.0,big-PI predictor and dbCAN3 softwares were used to predict and analyze the secreted proteins and carbohydrate active enzymes(C AZymes)of the protein sequence of M.hapla,and specific primer design and validation were carried out based on the gene sequence encoding the specific secretory protein.[Result]Among the 14 433 protein sequences of M.hapla,498 protein sequences had the characteristics of typical secretory proteins,accounting for 3.45%of the total protein sequences.The length of secretory proteins was mainly distributed in 100-300 amino acids;the signal peptide length of secreted proteins was mainly distributed in 17-27 amino acids.A total of 17 CAZymes were predicted from 498 secreted proteins,including glycoside hydrolases(GHs),polysaccharide lyases(PLs)and carbohydrate binding modules(CBMs).The M.hapla specific primer Mhs-F2/R2 could effectively discriminate M.hapla from M.enterolobii.[Conclusion]A total of 498 proteins sequences with typical secretory protein characteristics were predicted,and the length characteristics,amino acid composition and signal peptide length of secretory proteins were analyzed.Specific primers were designed based on the DNA sequence of the specific secretory protein(A0A1I8BG71)of M.hapla,which could distinguish M.hapla from M.enterolobii.
