STING抑制剂在脑卒中后炎症反应中的调控作用与研究进展
The regulatory role and research advances of STING inhibitors in post-stroke inflammation

Abstract
Stroke is an acute neurological injury caused by a sudden interruption of cerebral blood supply,which imposes a heavy burden on patients due to its high mortality and disability rates.Following a stroke,the cGAS-STING pathway becomes activated,strongly inducing a type Ⅰ interferon response that ex-acerbates stroke-related damage.Targeted inhibition or degra-dation of STING can alleviate excessive inflammatory signaling and improve therapeutic outcomes.Strategies for STING inhibi-tion include the use of covalent inhibitors and competitive an-tagonists that target the ligand-binding domain(LBD)to block cyclic dinucleotide(CDN)binding,thereby suppressing STING activation.In addition,selective degradation of STING via the ubiquitin-proteasome system has also demonstrated promising efficacy.This review examines the role of the cGAS-STING pathway in stroke and summarizes recent advances in the development of STING inhibitors and degraders for the treat-ment of stroke.We aim to provide new perspectives and strate-gies to support the development of innovative anti-stroke thera-pies targeting STING.关键词
