Scyllo-inositol mitigates blue-light-induced aging in Caenorhabditis elegans

Abstract
Aim To investigate the protective effects of scyllo-inositol on blue-light-induced aging in Cae-norhabditis elegans(C.elegans)and the underlying molecular mechanisms.Methods A blue-light irra-diation aging model was established.Synchronized L4 larvae were randomly assigned to a blank control group,a blue-light model group(blue-light intensity 22.0 cd·m-2,24 h exposure followed by 48 h recov-ery),Scyllo-inositol treatment groups(0.5,1,5 and 25 mmol·L-1 administered during blue-light expo-sure),and vitamin C positive-control groups(5 and 10 mmol·L-1 administered during blue-light expo-sure).Lifespan,locomotor capacity(head thrashing and body bending),reactive oxygen species(ROS)levels and lipofuscin autofluorescence were measured.Transcriptomic changes were analyzed by RNA-seq and validated by quantitative real-time PCR(qRT-PCR).Results Compared with controls,blue-light irradiation significantly shortened lifespan,reduced lo-comotor activity,increased ROS and lipofuscin levels,and disrupted pathways related to oxidative stress,lipid metabolism and xenobiotic detoxification.Scyllo-inositol treatment markedly prolonged lifespan(P<0.01),improved motility(P<0.05),reduced ROS(P<0.01)and lipofuscin accumulation(P<0.05),and partially restored the homeostasis of these pathways.qRT-PCR confirmed that scyllo-inositol reversed blue-light-induced aberrant expression of multiple antioxi-dant and metabolism-related genes.Conclusion Scyllo-inositol delays aging and extends lifespan in C.elegans by attenuating ROS accumulation,alleviating oxidative stress and immune over-activation,and re-establishing metabolic and detoxification homeostasis disrupted by blue-light irradiation.关键词
