和枢消积丸通过调控DUSP4/ERK表达抑制肝癌细胞生长的机制研究
Heshu Xiaoji prescription inhibits the growth of hepatocellular carcinoma cells by regulating the expression of DUSP4/ERK

Abstract
Objective:To explore the mechanism of Heshu Xiaoji prescription in inhibiting the growth of human hepatoma cell line Huh7.Methods:Different concentrations of Heshu Xiaoji prescription serum were used to intervene human hepatoma cell line Huh7,and CCK-8 colorimetric analysis was employed to evaluate the effect of Heshu Xiaoji prescription on Huh7 cell viability;Cell Scratch Test,Transwell migration and invasion test were used to evaluate the ability of migration and invasion;The apoptosis of Huh7 cells was detected by flow cytometry;The mRNA expressions of dual-specificity protein phosphatase 4(DUSP4),extracellular signal-regulated kinase(ERK)1 and ERK2 were detected by Real-time PCR;The expression of DUSP4,ERK1/2 and p-ERK1/2 protein was detected by Western blotting test.Results:Compared with the control group,Heshu Xiaoji prescription could effectively inhibit the proliferation,migration and invasion of Huh7 cells,and promote the apoptosis of Huh7 cells(P<0.05);Compared to the control group,all treatment groups exhibited upregulated DUSP4 mRNA levels along with downregulated ERK1/2 mRNA expression(P<0.05);Western blot analysis revealed that Heshu Xiaoji prescription significantly upregulated DUSP4 protein levels while suppressing both total and phosphorylated ERK1/2 expression(P<0.05).Conclusion:Heshu Xiaoji prescription can inhibit the proliferation,migration and invasion of tumor cells,and its anti-cancer effect may be related to the up-regulation of DUSP4 expression and the inhibition of ERK1/2 phosphorylation.关键词
