Abstract

[Objective]In order to study the effect of grazing on soil bacterial community in Hanas National Na-ture Reserve.[Method]Using 16S rRNA high-throughput sequencing technology,soil physical and chemical prop-erties analysis,plant diversity characteristics determination and correlation analysis,this study investigated the compo-sition and function of soil bacterial communities in grazing area(HG)and control area(CK).[Result](1)Com-pared with CK,the bulk density of HG was significantly increased,the plant coverage,plant height,soil water con-tent,available nitrogen and available phosphorus were significantly decreased,and the number of plant clusters and plant species were not significantly affected.(2)There were significant differences in the nine α diversity indexes rep-resented by Shannon_H and Chao_1 of soil bacteria between HG and CK,but there was no significant difference in β diversity.The abundance of Proteobacteria,Actinobacteria and Bacteroidetes in HG increased significantly(P<0.05).(3)There was a strong correlation between soil physical and chemical properties and soil bacterial community composition(P<0.05).Water content,available nitrogen,and available phosphorus were negatively correlated with representative genera such as Sphingomonas and Blastocatella in HG,but positively correlated with Gemmatimonas,Unidentified_eubacterium_EA25,and Candidatus_Solibacter in CK.Plant species richness,,coverage,and height were positively correlated with Unidentified_eubacterium_EA25 and Gemmatimonas in non-grazing areas(CK),and negatively correlated with Sphingomonas in grazing areas.[Conclusion]Grazing increased the diversity and abun-dance of soil bacterial community,which may mediate the composition of soil bacterial community through the changes of soil available nutrients and plant diversity chatacteristics.Due to the positive impact of moderate grazing on the diversity of soil bacterial communities,it is advisable to adopt varied grazing management approaches in pro-tected regions.This includes implementing seasonal rotational grazing in areas with lower soil nutrients and plant di-versity,while allowing moderate continuous grazing in meadows that exhibit high stress resistance.
