摘要

Abstract

In this study,to explore the genes related to peanut oil biosynthesis,a local peanut cultivar Huodou from Heyuan City,Guangdong Province,and a space-induced peanut mutant line(the 8th mutant generation,M8)were used as experimental materials.Compared with the original Huodo cultivar,the mutant line exhibited significant improvements in key yield-related traits(single-plant yield,total pods per plant,full pods per plant),while also displaying the superior agronomic characteristic of a compact plant architecture.Whole-genome resequencing was performed on both lines to conduct genetic variation analysis.By aligning against the reference genome(GCF_003086295),a range of genetic variations were systematically identified,including a total of 545416 single nucleotide polymorphisms(SNPs),167629 insertions and deletions(InDels),3032 copy number variations(CNVs),and 3525 structural variations(SVs).Among these variations,66 genes associated with peanut oil metabolism were screened out,including members of key gene families such as acyltransferase genes(DGATs)and SDP1 triglyceride lipase genes.Combined with quality trait evaluation,comparative analysis between the mutant line and the control revealed that the mutant exhibited significantly higher contents of crude fat(48.62%)and oleic acid(39.37%),whereas crude protein and linoleic acid contents were relatively lower.Functional annotation indicated that these candidate genes play important roles in the oil synthesis pathway.Notably,missense mutations in these genes may alter the conformation of the enzyme active center,thereby influencing the efficiency of triglyceride synthesis.These findings provide important target genes for subsequent functional validation studies and offer valuable references for exploring the molecular mechanisms underlying peanut space breeding.
