Abstract

To investigate the radiation biological effects of 60Co-γ rays on Prunus campanulata seeds,germinating seeds were exposed to graded doses(0,60,80,100,120 Gy)for a systematical analysis of dose-dependent impacts on germination,seedling growth,and physiological metabolism.Results demonstrated a significant negative correlation between radiation dose and relative germination rate(r=-0.740,P<0.01),with a semi-lethal dose(LD50)of 115.8 Gy calculated using the quadratic regression model y=-0.01 1x2+0.839 6x+100.27(R2=0.978 9).Radiation induced significant morphological alterations(P<0.05),including deeply lobed leaves,enhanced leaf serrations,and irregular leaf margins.Physiological analyses revealed dynamic responses:superoxide dismutase(SOD)activity initially increased and then decreased,peroxidase(POD)exhibited a V-shaped pattern,catalase(CAT)consistently remained suppressed across irradiated groups(P<0.05),malondialdehyde(MDA)content showed dose-dependent accumulation,and soluble sugar/protein levels displayed biphasic trends(initial decline followed by recovery).Based on integrated growth and physiological metrics,the optimal radiation dose range was determined to be 60-100 Gy.This study pioneers a radiation dose-effect model for Prunus campanulata,providing critical theoretical and practical guidance for radiation mutagenesis breeding in Prunus.
