Response Surface Optimization of Enzyme-Assisted Ultrasonic Extraction of Anthocyanins from Vaccinium vitis-idaea L.

Abstract
Taking the fruits of Vaccinium vitis-idaea L.as the raw material and the anthocyanins extraction yield as the evaluation index,single-factor experiments were conducted to evaluate the effects of enzyme type,ultrasonic time,ultrasonic temperature,ethanol concentration,enzyme dosage,enzymatic hydrolysis temperature,enzymatic hydrolysis time,and solid-to-liquid ratio on the anthocyanins extraction yield.Response surface methodology(RSM)was used to optimize the extraction process.The results showed that the anthocyanin extraction yield of the cellulase-treated group was 11.5%and 7.6%higher than that of the non-enzyme control group and pectinase group,respectively.The basic process conditions optimized by single-factor experiments were as follows:cellulase dosage of 1%,enzymatic hydrolysis temperature of 35℃,enzymatic hydrolysis time of120min,and solid-to-liquid ratio of 1∶20 g·mL-1.The optimal process optimized by RSM was determined as follows:ultrasonic time of 21.69 min,ultrasonic temperature of 39.67℃,and ethanol concentration of 51.95%Under these conditions,the predicted anthocyanin extraction yield was 3.54 mg·g-1,and the actual verified extraction yield was 3.53±0.02 mg·g-1,which was in good agreement with the predicted value.The cellulase-assisted ultrasonic extraction process established in this study provides a theoretical basis and technical support for the efficient extraction of anthocyanins from Vaccinium vitis-idaea L.cellulase-assisted ultrasonic extraction technology for anthocyanins from Vaccinium vitis-idaea L.,which provides a theoretical and technical foundation for the in-depth development and utilization of anthocyanins.关键词
