甘草泻心汤加减对小鼠口腔溃疡治疗作用及其主要成分相关性研究
Therapeutic Effects of Modified Gancao Xiexin Decoction on Oral Ulcers in Mice and Correlation Analysis of its Main Components

Abstract
Objective:To investigate the therapeutic effects of modified Gancao Xiexin Decoction on oral ulcers in mice and to ana-lyze the correlation between its chemical components and ulcer healing.Methods:The modified Gancao Xiexin Decoction was ex-tracted using water extraction,water extraction followed by alcohol precipitation,ethanol reflux,and water extraction followed by flocculation.An oral ulcer model was established in mice to compare the effects of extracts obtained by different methods on ulcer healing.Histological sections were used to observe morphological changes in ulcer tissues,and serum immune-related factors were measured by enzyme-linked immunosorbent assay(ELISA).The main chemical components of the extracts were analyzed using liq-uid chromatography-high-resolution mass spectrometry(LC-HRMS).According to the 2020 edition of the Chinese Pharmacopoeia,19 typical compounds from each herb were identified,and their contents in the extracts were quantified.Partial least squares regres-sion(PLSR)analysis was used to examine correlations between these compounds,body weight gain in mice,and ulcer healing.Re-sults:Extracts from both the original and modified Gancao Xiexin Decoction accelerated oral ulcer healing and reduced weight loss in mice.Among the different extraction methods,the water extraction followed by flocculation showed the best effect.Serum granulo-cyte-macrophage colony-stimulating factor(GM-CSF)levels increased from 578 pg/mL to 1 214 pg/mL during the early stage of ul-cers and decreased to 661 pg/mL after healing.LC-MS identified a total of 66 small molecular compounds.PLSR analysis indicated that naringenin,taraxasterol,and baicalein were most strongly correlated with body weight gain and ulcer recovery.Conclusion:Modified Gancao Xiexin Decoction promotes oral ulcer healing,with the water extraction followed by flocculation method being the most effective.The healing effect is likely due to the combined action of multiple chemical components in the medicinal materials.关键词
