PGE2对人子宫内膜间质细胞迁移的影响及作用机制实验研究
Effect of PGE2 on the migration of human endometrial stromal cells and its mechanism

Abstract
Objective:To investigate the effect of prostaglandin E2(PGE2)on the migration of human endome-trial stromal cells(hESCs)and its possible mechanism.Methods:hESCs were randomly divided into a control group(treated with an equal volume of medium without other intervention),a PGE2 group(treated with 25 μmol/L PGE2),an oxytocin group(treated with 10 nmol/L oxytocin),a PGE2+NOD-like receptor family pyrin domain-con-taining protein 3(NLRP3)inhibitor group(treated with 25 μmol/L PGE2+10 μmol/L MCC950,an inhibitor of NLRP3),and a PGE2+Caspase-1 inhibitor group(treated with 25 μmol/L PGE2+20 μmol/L VX-765,an inhibitor of Caspase-1).The level of PGE2 in hESCs was detected by ELISA.The cell migration rate was measured by wound-healing assay.The mRNA expressions of NLRP3,cleaved Caspase-1,interleukin-1β(IL-1 β),matrix metalloprotein-ase-2(MMP-2),and MMP-9 were determined by RT-qPCR.The protein expressions of NLRP3,cleaved Caspase-1,IL-1β,MMP-2,and MMP-9 were detected by Western blot.Results:The cell migration rate,intracellular PGE2 level,and mRNA and protein expressions of NLRP3,cleaved Caspase-1,IL-1β,MMP-2,and MMP-9 in the PGE2 group were higher than those in the control group and the oxytocin group,and those in the oxytocin group were higher than those in the control group(all P<0.05).The above indicators in the PGE2+NLRP3 inhibitor group and the PGE2+Caspase-1 inhibitor group were lower than those in the PGE2 group(all P<0.05).Conclusion:PGE2 can promote the migration of hESCs,and its mechanism may be related to the activation of the NLRP3/Caspase-1/IL-1β pathway.关键词
