New Strategy for Pretreatment of Hydrated Soft Biological Samples for SEM:a Simple and Cost-effective Approach Based on RTILs

Abstract
To demonstrate the utility of the RTIL-based SEM pretreatment method for hydrated soft biological specimens,this study replaced the conventional time-consuming,labor-intensive,and expen-sive SEM sample preparation with a simple droplet application and blotting treatment.The results showed that although the RTIL-based method did not achieve perfect preservation of morphology,it produced results comparable to those of conventional methods.Specifically,it demonstrated superior structural fidelity in preserving spore quantity,spore chains,leaf trichomes,and epidermal cells,provi-ding a more realistic representation of biological structures than traditional SEM pre-treatment.Addi-tionally,its electrical conductivity was comparable to that of conventional methods.The developed RTIL-based approach shows great potential as an alternative to conventional SEM pretreatment,offer-ing advantages in cost,simplicity,and efficiency,while maintaining image quality nearly identical to that of traditional protocols.Notably,it outperforms standard methods in preserving critical structural features such as the bead-like morphology of Alternaria spore chains and cellular turgor.关键词
