Abstract

This study was aimed at developing monoclonal antibodies(mAbs)to the nucleoprotein of Dabie bandavirus and identi-fying a specific mAb pair suitable for sandwich-based antigen detection with immunocolloidal gold,immunofluorescence chromatogra-phy,and enzyme-linked immunosorbent assay(ELISA).The recombinant nucleoprotein was expressed in HEK293 suspension cells,purified,and used to immunize BALB/c mice.Hybridoma technology was then used to generate mAbs.After immunization,mouse se-rum at a dilution of 1:10 000 showed strong antigen-antibody reactivity,with ELISA OD450 values ranging from 1.38 to 2.56.Through systematic screening,65 hybridoma cell lines were obtained,and one specific mAb pair with high specificity was successfully identi-fied.This antibody pair demonstrated positive binding to Dabie bandavirus antigen in immunocolloidal gold test strips,fluorescence immunochromatographic strips,and ELISA.The results confirmed the successful development of a practical mAb pair that can be ef-fectively applied for the detection of Dabie bandavirus antigen in multiple immunological assays.
