基于D-loop和Cyt-b基因的黑线姬鼠种群遗传结构与汉坦病毒感染关联性分析
Association between Apodemus agrarius population genetic structure,according to the D-loop and Cyt-b genes,and hantavirus infection

Abstract
This study was aimed at elucidating the genetic structure of Apodemus agrarius populations in Hunan,China,and its association with hantavirus infection.Total DNA extracted from rodent lung tissues served as the template for PCR amplification of con-served mitochondrial D-loop and Cyt-b gene regions,and was followed by sequencing.Sequence characteristics and genetic diversity were analyzed in MEGA 6 and DnaSP 5 software,and phylogenetic trees were constructed with the neighbor-joining(NJ)and maxi-mum likelihood(ML)methods to assess population differentiation and virus-host linkages.Both genes showed AT-rich composi-tions[60.9%for D-loop(658 bp);60.7%for Cyt-b(871 bp)].A total of 38 and 40 variable sites were detected,corresponding to 11 and 9 haplotypes.The haplotype diversity(H)values were 0.956 and 0.912,and the nucleotide diversity(π)was 0.012 78 and 0.009 51.Tajima's D values were-1.446 26 and-1.555 71.Phylogenetic analysis indicated that most Liuyang populations(except LY-029-HTAA-7 and LY-031-HTAA-9)formed a monophyletic clade,whereas the Shuangfeng population(SF-014-HTAA-7)clustered in-dependently.Notably,an uninfected sample(LY-03-UAA-6)and an infected sample(LY-016-HTAA-6)were closely grouped in the Cyt-b tree.This study revealed that A.agrarius populations in Hunan exhibited notable genetic diversity,and no direct correlation was observed between hantavirus infection and mitochondrial genome variation.Our findings highlight the need to integrate host eco-logical behavior and environmental factors in future studies to elucidate virus transmission mechanisms.关键词
