1株麦氏弧菌噬菌体vB_VmeS_MHP的分离鉴定及其基因组特征分析
Isolation,identification,and analysis of genomic characteristics of the Vibrio metschnikovii phage vB_VmeS_MHP

Abstract
This study developed novel strategies for controlling Vibrio metschnikovii infections and phage-based formulations.Mi-gratory bird-derived Vibrio metschnikovii VM4F served as the host strain.Lytic phages were isolated and purified from lake water samples collected from the host habitat via the double-layer plate method.Subsequently,biological characterization and whole-genome bioinformatics analysis were conducted on the isolated phages.A Vibrio metschnikovii-specific lytic phage was successfully ob-tained and designated as vB_VmeS_MHP.The optimal multiplicity of infection(MOI)of this phage was determined to be 0.001,its lytic cycle was measured as 110 minutes,and the burst yield was calculated to be 404 PFU/cell.This phage demonstrated strong stress tolerance:it withstood heat at 60 ℃,acidic/alkaline conditions ranging from pH 5 to 12,and 235.7 nm UV irradiation for 90 minutes.In vitro bacteriostatic activity persisted for 6 hours,and it exhibited significant biofilm-disrupting effects.Morphological observations revealed that the phage had a head diameter of(59.3±1.8)nm and a tail length of(207.5±0.5)nm.The phage was classified within the family Siphoviridae of the order Caudovirales.Whole-genome analysis indicated that the phage possessed a linear double-stranded DNA genome with a length of 41 255 bp and a GC content of 43.48%.No virulence or antibiotic resistance genes were detected in its genome.Phylogenetic analysis suggested shared evolutionary relationships between this phage and phages from multiple regions and hosts,thus implying that cross-host jumping and recombination-driven evolution might have occurred.This study confirmed that vB_VmeS_MHP possesses excellent antibacterial efficacy and environmental adaptability,and it provides crucial theoretical support for the development of Vibrio metschnikovii phage-based control products.关键词
