Abstract
Intensive aquaculture,as the primary production method in modern aquaculture,has significantly increased economic efficiency per unit water volume.However,this high-density farming model has also resulted in severe nitrogen pollution.To screen aerobic denitrifying bacterial strains suitable for aquaculture water treatment,this study employed a comprehensive screening approach and isolated a highly efficient aerobic denitrifying bacterium from pond sediment.Preliminary identification based on morphological observation and 16S rDNA gene sequence analysis confirmed the strain as Alcaligenes ammonioxydans,designated C-4.Subsequent investigations focused on its nitrogen removal capacity,biosafety,and application efficacy.The results demonstrated that under monoculture conditions(inoculum size:1%,C/N=10,temperature:30℃,agitation speed:200 r/min),strain C-4 achieved 100%NO3--N removal and 60.83%NH4+-N removal.Under mixed nitrogen source conditions,strain C-4 exhibited 100%NO3--N removal,69.48%NH4+-N removal,and 87.92%total nitrogen(TN)removal.Safety evaluation confirmed the strain's non-hemolytic activity and lack of toxicity to grass carp(Ctenopharyngodon idella),indicating high biosafety.In simulated aquaculture wastewater trials,strain C-4 exhibited(i)(90.97±0.08)%NO3--N removal;(ii)(61.35±0.15)%NH4+-N removal;(iii)(46.90±0.14)%NO2--N removal;and(iv)(77.31±0.01)%TN removal.Collectively,these findings highlight strain C-4 as a promising candidate for aerobic denitrification in aquaculture water purification applications,owing to its high nitrogen removal efficiency and demonstrated environmental safety.关键词
