Abstract

Sodium dehydroacetate(Na-DHA)has been reported to induce coagulation disorders in livestock and poultry by inhibiting the vitamin K cycle and to impair skeletal development in fetal rodents.To investigate the effects of Na-DHA on osteoblasts and bone metabolism indicators,chicken bone marrow mesenchymal stem cells(BMSC)were isolated,cultured,and induced to differentiate into osteoblasts.The differentiated osteoblasts were treated with different concentrations of Na-DHA(0,0.2,0.5,and 1.0 mmol/L),and the effects of Na-DHA on osteoblast mineralization,the vitamin K cycle,and bone metabolism were evaluated.The results showed that chicken osteoblasts were successfully induced.Alkaline phosphatase(ALP)staining demonstrated that ALP activity was significantly decreased in the 0.5 and 1.0 mmol/L Na-DHA groups compared with the Control group(P<0.05 or P<0.01).Alizarin red S staining revealed a significant reduction in the number of mineralized nodules in the 0.5 and 1.0 mmol/L Na-DHA groups compared with the Control group(P<0.05 or P<0.01).In addition,compared with the Control group,intracellular vitamin K content in chicken osteoblasts were markedly reduced in the 0.5 and 1.0 mmol/L Na-DHA groups(P<0.001).The protein expression levels of vitamin K epoxide reductase complex subunit 1(VKORC1)(P<0.01 or P<0.001)and vitamin K epoxide reductase complex subunit 1-like protein 1(VKORC1L1)(P<0.05)were significantly decreased.Moreover,the protein expression levels of bone gamma-carboxyglutamate protein(BGP)and osteoprotegerin(OPG)were significantly reduced(P<0.05 or P<0.01),whereas the expression level of receptor activator of nuclear factor-κB ligand(RANKL)was significantly increased(P<0.05).These results indicate that Na-DHA impairs the mineralization of chicken osteoblasts by inhibiting the vitamin K cycle and disrupting bone metabolism.The findings of this study provide a reference for evaluating the potential adverse effects of Na-DHA on the skeletal system of poultry.
