加味青娥丸对绝经后骨质疏松症患者血清外泌体miRNA表达谱的影响
Effect of modified Qing'E formula on miRNA expression profile of serum exosomes in patients with postmenopausal osteoporosis

Abstract
Objective To investigate the effect of modified Qing'E formula(MQEF)on the miRNA expression profile in serum exosomes of postmenopausal osteoporosis(PMOP)patients and to identify miRNAs showing a significant inverted"V"-shaped trend.Methods According to strict inclusion and exclusion criteria,6 healthy postmenopausal women were enrolled as the healthy control group and 12 PMOP patients(marked as PMOP group before treatment)were treated with MQEF for three months(marked as MQEF group after treatment).Serum samples were collected from the three groups,and serum exosomal miRNAs were extracted.The DNBSEQ sequencing platform was used for data filtering and quality control.Differential analysis and expression ranking of the obtained miRNAs were performed,and qRT-PCR was used to validate the sequencing results.Bioinformatics analysis was conducted to identify the main targets and related pathways of MQEF.Results A total of 1 130 miRNAs were extracted from the three groups.There were 54 significantly differentially expressed miRNAs(1 downregulated and 53 upregulated)in serum exosomes between the healthy control group and the PMOP group,of which the top five were hsa-miR235-3p,hsa-miR26-5p,hsa-miR239-5p,hsa-miR121-5p and hsa-miR148-3p.Between the PMOP group and the MQEF group,there were 11 significantly differentially expressed miRNAs(10 downregulated and 1 upregulated),of which the top five were hsa-miR177-3p,hsa-miR148-3p,hsa-miR294-3p,hsa-miR162-3p and hsa-miR260-5p.Among them,has-miR148-3p not only showed significant differential expression across the three groups but also exhibited an inverted"V"-shaped trend in expression levels.qPCR validation results showed that hsa-miR148-3p was upregulated 7.7-fold in the PMOP group and downregulated 9.9-fold in the MQEF group,with significantly statistical differences(P<0.05).A total of 212 predicted target genes of hsa-miR148-3p related to PMOP were identified.PPI protein network interaction analysis suggested that its potential targets might be STAT3,SRC and TGFB1.Enrichment analysis was performed on the significantly differentially expressed miRNAs following MQEF treatment.GO enrichment analysis demonstrated that MQEF treatment for PMOP was associated with key processes inclu-ding enzymatic catalysis and oxidative stress.KEGG enrichment analysis revealed that MQEF treatment for PMOP was closely related to the mTOR signaling pathway.Conclusion MQEF treatment for PMOP may be related to the mTOR sig-naling pathway and oxidative stress.MQEF can significantly downregulate the level of miR148-3p in serum exosomes of PMOP patients and may exert its effects through potential targets such as STAT3,SRC and TGFB1.关键词
