摘要

Abstract

AIM To optimize the extraction process for total flavonoids from Eucommiae folium Oliv.leaves,and to evaluate their anti-oxidant activity.METHODS With ethanol concentration,liquid-solid ratio,soaking time and extraction time as influencing factors,the comprehensive score for total flavonoids,rutin,isoquercitrin,astragalin,phlorizin,quercetin,kaempferol contents and extract yield as evaluation index,the anti-oxidant activity was optimized by AHP-entropy weight TOPSIS method combined with Box-Behnken response surface method.Subsequently,the scavenging rates of DPPH·,ABTS+·free radicals were determined.RESULTS The optimal conditions were determined to be 60%for ethanol concentration,21∶1 for liquid-solid ratio,1.5 h for soaking time,2.0 h for extraction time,and two times for extraction frequency,the various constituent contents and extract yield were 79.671,0.131,1.127,0.035,2.143,0.855,0.157 mg/g and 37.534%.The IC50 values of total flavonoids on DPPH·,ABTS+·free radicals were 0.028,0.053 mg/mL,respectively.CONCLUSION This stable and reliable method can be used for the extraction of total flavonoids from E.folium leaves with strong anti-oxidant activity.
