基于转录组学技术探究葛根多糖缓解热应激致肉鸡肝脏损伤的保护作用及机制
Protective Effects and Mechanisms of Pueraria lobata Polysaccharides in Alleviating Heat Stress-Induced Liver Injury of Broilers Based on Transcriptomics Technology

Abstract
This experiment aimed to investigate the protective effects and mechanisms of Pueraria lobata poly-saccharides in alleviating heat stress-induced liver injury of broilers based on transcriptomics technology.A total of 120 one-day-old Arbor Acres(AA)white feather broilers were reared to 21 days of age,then randomly di-vided into 4 groups with 6 replicates per group and 5 broilers per replicate.Broilers in control group(NC group)and heat stress group(HS group)were fed basal diets,and others in experimental groups were fed basal diets supplemented with 100(PLP-L group)and 400 mg/kg(PLP-H group),respectively.Broiler chickens in HS group,PLP-L group,and PLP-H group were subjected to heat stress treatment at 21 to 35 days of age.The experiment lasted for 35 days.The results showed as follows:1)compared with NC group,the fi-nal body weight(FBW),average daily gain(ADG),average daily feed intake(ADFI)and liver superoxide dismutase(SOD),catalase(CAT)activities of HS group were significantly decreased(P<0.05),and the serum aspartate aminotransferase(AST),alanine aminotransferase(ALT)activities and liver malondialde-hyde(MDA),reactive oxygen species(ROS),interleukin-6(IL-6),interleukin-1β(IL-1β),tumor necro-sis factor-α(TNF-α)contents and heat shock protein 70(HSP70),heat shock protein 90(HSP90)protein expression levels were significantly increased(P<0.05).2)Compared with HS group,the serum AST,ALT activities and liver ROS content and HSP70,HSP90 protein expression levels of PLP-L group and PLP-H group were significantly decreased(P<0.05);the FBW,ADG and liver SOD activity of PLP-L group were significantly increased(P<0.05),and the liver MDA,IL-6,IL-1β and TNF-α contents were significantly de-creased(P<0.05).3)The transcriptomics analysis results indicated that the core genes involved in the action of Pueraria lobata polysaccharides included 17-beta-hydroxysteroid dehydrogenase 4(4HSD17B4)and 3-hydroxyacyl CoA dehydrogenase(EHHADH)et al,and the core pathways were mainly concentrated in perox-isome proliferator-activated receptors(PPARs),fatty acid metabolism and fatty acid degradation signaling pathways.In conclusion,the Pueraria lobata polysaccharides can alleviate the weight loss of broilers caused by heat stress,improve growth performance,antioxidant capacity and anti-inflammatory level,and improve liver tissue structure;the mechanism may be by regulating PPARs,fatty acid metabolism and fatty acid degradation signaling pathways and mediating the expression of related genes.[Chinese Journal of Animal Nutrition,2026,38(5):3387-3402]关键词
