Effect of Menghe Medical School's Characteristic Processed Wine-washed Angelica Sinensis on Femoral Head Microstructure and Related Signaling Pathways in Rats with Early-stage Osteonecrosis of the Femoral Head

Abstract
Objective:To explore the influence of Menghe medical school's characteristic processed wine-washed angelica sinensis on femoral head microstructure,the levels of OPG,RANKL,RANK,Wnt1 and Wnt3A,and related signaling pathways in rats with early-stage osteonecrosis of the femoral head(ONFH).Methods:Eighteen rats with ONFH were randomly divided into three groups(n=six per group):the early-stage ONFH model group,the model+traditionally processed wine-baked Angelica sinensis group,and the model+Menghe medical school's characteristic processed wine-washed Angelica sinensis group.Rats in the early-stage ONFH model group were administered normal saline by gavage,those in the model+traditionally processed Angelica sinensis group were given traditionally processed Angelica sinensis(4 mg·kg-1)by gavage,and those in the model+Menghe medical school's characteristic processed wine-washed Angelica sinensis group were given Menghe medical school's characteristic processed wine-washed Angelica sinensis(4 mg·kg-1)by gavage.All groups received consecutive interventions for 30 days.Following the intervention,all rats were euthanized.Femoral bone thickness and bone mineral density(BMD)were evaluated by X-ray examination.Morphological changes in the femoral head bone tissue were observed using hematoxylin-eosin(HE)staining.The expression levels of OPG,Wnt1,Wnt3A,RANK,and RANKL in the femoral head bone tissue were detected by immunohistochemistry.Results:X-ray findings of the femurs showed that in the model group,there were multiple high-density sclerotic areas of various morphologies within the femur,along with increased bone thickness and a marked increase in BMD.In the model+traditional processed Angelica sinensis group and the model+Menghe medical school's wine-washed Angelica sinensis group,bone thickness and BMD were reduced.HE staining showed that the model group exhibited articular cartilage erosion,an uneven cartilage surface,absence of the superficial layer,and a disrupted tidemark.The model+traditional processed Angelica sinensis group showed articular cartilage damage,an irregular cartilage surface,and an incomplete tidemark,while the model+Menghe medical school's wine-washed Angelica sinensis group exhibited mild articular cartilage damage,a relatively smooth cartilage surface,and a relatively intact tidemark.Compared with the model group,both treatment groups(model+traditional processed Angelica sinensis and model+Menghe medical school's wine-washed Angelica sinensis)showed significantly increased protein expression levels of OPG,Wnt1,and Wnt3A,and significantly decreased protein expression levels of RANK and RANKL in the femoral head bone tissue(P<0.05).Furthermore,the improvement in these indicators was significantly better in the Menghe medical school's wine-washed Angelica sinensis group than in the traditional processed Angelica sinensis group(P<0.05).Conclusion:Menghe characteristic processed Angelica sinensis may treat ONFH by modulating the expression of proteins involved in the NF-κB,OPG/RANKL/RANK,and Wnt/β-catenin signaling pathways,thus suppressing inflammation and immune responses.关键词
